As an example, in the case of interstitial lung disease (ILD). ILD is a severe complication of rheumatoid arthritis, often challenging to manage due to limitations in current treatments. JAK inhibitors (JAKi) have demonstrated significant potential; however, they are limited by challenges including low bioavailability, a short duration of action, and their characterization as P-glycoprotein substrates. This project aims to develop advanced drug delivery carriers, including biomimetic camouflaged nanoparticles, to enhance therapeutic outcomes. Nanoparticles loaded with both JAKi and PGPi and coated with membranes derived from platelets and A549 cells will target inflamed pulmonary environments, minimizing systemic side effects and improving drug specificity. Inhalable formulations, such as liquid aerosols and dry powders, will be explored to ensure efficient lung delivery. Comprehensive evaluations, including <em>in vitro<\/em> studies, <em>ex vivo<\/em> models and deep lung simulations with NGI impactors and breathing simulators, will validate the approach.
